Metarhizium Anisopliae
Metarhizium anisopliae is a fungus that grows upon insect host cuticle. The main enzyme involved in this growth appears to be a subtilisin-like serine protease [1] that initiates protein degradation. This is the major protein secreted into the host cuticle during invasion [2]. This protease activity is then followed by the action of exopeptidases such as carboxypeptidases, which release individual amino acids of the host for nutritional utilization. Although fungi typically contain serine carboxypeptidase activity, a novel carboxypeptidase was discovered in Metarhizium anisopliae by St. Leger et al. [3] that was inhibited by 1,10-phenanthroline. However, it did not appear to be a true metallocarboxypeptidase because the active site serine was blocked by DipF.
Joshi & St. Leger [4] succeeded in identifying a true fungal metallocarboxypeptidase that they designate by the abbreviation MeCPA. Discovery was based on cloning RNA from genes expressed when the fungus was grown on insect cuticle. Ten thousand plaques were screened and 10 were positive. The largest of these provided the sequence for MeCPA. This is the first fungal metallocarboxypeptidase to be characterized.
The entomopathogenic fungus Metarhizium anisopliae (Metschn.) Sorokin is widely used for biocontrol of pest insects, and many commercial products are on the market or under development. The aim of this review is to summarise all relevant safety data of this fungus, which are necessary for the commercialisation and registration process. The review contains the following sections: (1) identity, (2) biological properties (history, natural occurrence and geographical distribution, host range, mode of action, production of metabolites/toxins, effect of environmental factors), (3) methods to determine and quantify residues, (4) fate and behaviour in the environment (mobility and persistence in air, water and soil), (5) effects on non-target organisms (microorganisms, plants, soil organisms, aquatic organisms, predators, parasitoids, honey bees, earth worms, etc.), (6) effects on vertebrates (fish, amphibia, reptiles, and birds), and (7) effects on mammals and human health (allergy, pathogenicity/toxicity). On the basis of the presented knowledge, M. anisopliae is considered to be safe with minimal risks to vertebrates, humans and the environment.
